“Investigating the immune microenvironment of primary estrogen receptor positive (ER+) breast cancer”

Objective:
- Immunotherapy has revolutionized outcomes in several different tumor types but is relatively unsuccessful in treating ER+ breast cancer. Our project aims to dissect the immune microenvironment through multipronged approaches for assessing the type and function of infiltrating immune cell subsets in treatment-naive tumor tissue and peripheral blood from patients with ER+ invasive ductal carcinoma (IDC) and invasive lobular carcinoma (ILC), the two major histological subtypes of ER+ breast cancer. Through comprehensive global analysis of immune cells and their potential interactions, we identified macrophages and CD8 T cells as important subsets that can be potentially targeted in ER+ breast cancer to improve patient outcomes.

Objective:
- The ongoing COVID-19 pandemic, caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), led to more than 126,265,724 million infected individuals and over 2,769,853 global deaths by the end of March 2021. The novel coronavirus SARS-CoV-2 is an enveloped, positive-sense, single-stranded RNA betacoronavirus of the family Coronaviridae. The main target of these virus is the lungs, but new reports about COVID-19 indicate its effects on other organs, including kidney, brain and testis. SARS-CoV-2 uses the angiotensin-converting enzyme 2 (ACE2) receptor and the transmembrane protease serine 2 (TMPRSS2) to invade human cells. Based on ACE2 and TMPRSS2 expression, the testis has been classified as being at high risk of infection by SARS-CoV-2. Indeed, patients with COVID-19 suffer from testicular discomfort, which can be associated with viral orchitis. Similarly, some studies have reported reduced testosterone levels in the serum from COVID-19 patients. These data suggest that COVID-19 could affect male reproductive function, in fact the presence of SARS-CoV-2 has been detected in the testis of some deceased patients of COVID-19 as well as in some semen samples of patients in the acute and recovery phase of COVID-19. Our preliminary analysis of the testis of 5 patients deceased due to COVID-19 and their respective controls showed increased germ cell apoptosis, micro thrombosis, and infiltration of lymphocyte CD3+. Strikingly, we observed that almost all the apoptotic cells were spermatogonial stem cells (SSCs). Non-apoptotic SSCs also displayed signs of DNA damage and oxidative stress. Using single nuclei RNA-seq, RNAscope, and immunohistochemistry, we will interrogate the effects of COVID-19 on human testis from the macroscopic presentation to the single cell, providing an important basis to understand the SSC alterations that may hamper fertility in COVID-19 survivors.

The Cytometry Facility is proud to announce the Cytoflex LX is on the way! For our last virtual seminar, we will be re-introducing the technology. In brief, the CytoFLEX LX Flow Cytometer is an expansion of the CytoFLEX Platform. This flagship model expands research possibilities with six lasers and 21 color parameters. Six spatially separated lasers allow panels to be spread across the spectrum reducing cross talk and spectral overlap. The fully activated instrument includes three fluorescent channels from the 355 nm (UV) laser, five from the 405 nm (Violet) laser, three from the 488 nm (Blue) laser, five from the 561 nm (Yellow Green) laser, three from the 638 nm (Red) laser, and two from the 808 nm (Infrared) laser. The available 96-well plate sample loader allows for high-throughput applications and fits inside the instrument preserving the compact footprint. This event will start at 9:00 a.m. with numerous talks from trainees and conclude with a keynote lecture at 12:30 p.m.
Our Keynote speaker is Dr. Graham Walker, American Cancer Society Professor and HHMI Investigator at MIT, and a member of the National Academy of Sciences. He will deliver a talk from 12:30 to 1:30 p.m. entitled “Unravelling Mutagenic Translesion Synthesis: A Journey from the Ames Test to Cancer Chemotherapy,” and will meet with trainees afterward. Dr. Walker holds an R35 NIEHS merit award entitled, “Mechanisms of Eukaryotic Environmental Mutagenesis” and is researching therapeutics that inhibit mutagenic translesion DNA synthesis to sensitize cancer cells to chemotherapy.

The Institute for Precision Medicine hosts a monthly lecture series to highlight advances in precision medicine research and personalized care both at Pitt/UPMC and nationwide. This month, we are excited to host Amanda Paulovich, MD, PhD as a guest speaker. Dr Paulovich is the Aven Foundation Endowed Chair, and Director, CLIA Targeted Proteomic Laboratory at Fred Hutchinson Cancer Research Center. She also serves as the Director of the Clinical Research Proteomics Platform at the Brotman Baty Institute for Precision Medicine and is Professor of Medicine in the divison of Oncology at the Unviersity of Washington School of Medicine.
Dr. Paulovich received her M.D. and Ph.D. with Dr. Lee Hartwell at the University of Washington. She completed a residency in Internal Medicine at Massachusetts General Hospital and a fellowship in Oncology at Dana-Farber/Partners Cancer Center in Boston. She completed postdoctoral training at MIT-Whitehead Center for Genomics Research working with Dr. Eric Lander. As an oncologist, Dr. Paulovich was struck by the paucity of quantitative assays for measuring clinically relevant phenotypes in her patients, and the limitations that this put on her ability to practice “personalized medicine.” Out of these experiences, she became passionate about developing technologies and strategies for translation of novel diagnostics and therapeutics to enable precision medicine. Over the past 18 years, Dr. Paulovich’s research has focused on relieving a roadblock in biomedical research: a lack of validated and standardized tools for reliably quantifying human proteins. She has developed quantitative proteomic assays for measuring immunotherapy response, DNA damage response and growth factor receptor signaling to name a few. She was inducted to American Society for Clinical Investigation, received the 2014 Life Science Innovation Northwest Women to Watch in Life Science Award, received the 2015 Distinguished Achievement in Proteomic Sciences Award from the Human Proteome Organization, was awarded the Aven Foundation Endowed Chair in 2018, and was appointed Director of the Clinical Research Proteomics Platform of the Brotman Baty Institute for Precision Medicine in 2019.
In her presentation Dr Paulovich will discuss the added value of proteogenomics to the current genome-driven approach to precision oncology, and summarize the growing incorporation of targeted proteomic measurements based on selected/multiple reaction monitoring (SRM/MRM) mass spectrometry into clinical trials and the clinical laboratory. The establishment of proteogenomics represents an important component towards implementation of broader precision medicine efforts in cancer and beyond. The Bridgeside Research Forum is a seminar given by postdocs and graduate students every Friday from 12-1 pm during the academic year. Most weeks consist of two 30-minute talks, but there will be an occasional hour-long special seminar from an invited guest too.
The goal of this forum is to provide an opportunity for trainees to gain valuable experience presenting their research to a broad scientific community. The topics are diverse each week, so there is sure to be something for everyone. All are welcome to attend and enjoy a free lunch while you are there!
There is a free shuttle to transport people directly from the Hillman Cancer Center to Bridgeside Point II and back. The shuttle will depart Hillman at 11:30 am and then depart Bridgside at 1:15 pm
