| Pitt Hosts Vaccine Film Premiere April 12 Online Panel discussion follows “Chasing COVID,” a 30-minute film featuring the work of Pitt’s Center for Vaccine Research (CVR), will be available online at 7 p.m. on Monday, April 12, the 66th anniversary of the Salk polio vaccine being declared “safe and effective.” Created by filmmaker and Pitt senior lecturer Carl Kurlander, this engaging program takes viewers inside the lab of CVR director Paul Duprex, PhD, who, with his team, is engaged in a global collaboration to develop a COVID-19 vaccine that encompasses what is known about the virus’ emerging variants. In addition to Duprex and Kurlander, a post-screening panel discussion will feature: · Peter Salk, MD, professor of infectious diseases and microbiology at the Graduate School of Public Health and president of the Jonas Salk Legacy Foundation. One of the first to get his father’s then-experimental polio vaccine, he will reflect on lessons that historic collective effort has in today’s battle against COVID-19. · Sylvia Owusu-Ansah, MD, MPH, assistant professor of pediatrics and of emergency medicine. One of the earliest African American physicians to be vaccinated against COVID-19, she will talk about her engagement with communities concerned about vaccine hesitancy and equitable vaccine distribution. · Anita McElroy, MD, PhD, assistant professor of pediatrics. In addition to collaborating with Duprex on a measles-vector vaccine candidate for COVID-19, McElroy runs the F*A*B Lab, which focuses on some of the world’s deadliest infectious diseases such as Ebola and Rift Valley fever virus. · The panel will be moderated by Kornelia Tancheva, director of the University of Library System.

“In depth single cell profiling of a case of bone metastases with associated organoid models reveals a precision medicine approach to treatment”

Objective:
- Occurring in 65-80% of metastatic breast cancer (BC), bone metastasis (BoM) is the major cause of BC related mortality. Like other cancers, BC BoM has high inter- and intra-patient heterogeneity. Thus, understanding disease evolution and heterogeneity of BoM at the individual patient level will be key to guide precise application of targeted therapies. In this study, we described in depth histologic and single cell molecular characterization of a case of invasive lobular breast cancer (ILC) metastasis to bone, and examined potential therapeutic targets with associated organoid models.
“Zinc protection of fertilized eggs is an ancient feature of sexual reproduction in animals”

Objective:
- In most sexually reproducing organisms, it is vital that the egg is fertilized by only one sperm. In order to ensure this occurs, eggs utilize polyspermy blocks, mechanisms that inhibit the entry of multiple sperm. Recently, it has been observed that mammalian eggs release zinc at activation, a phenomenon hypothesized to contribute to the polyspermy block. We seek to demonstrate that this zinc release is conserved in non-mammalian eggs, and to directly test zinc’s ability to inhibit fertilization.

“Investigating the immune microenvironment of primary estrogen receptor positive (ER+) breast cancer”

Objective:
- Immunotherapy has revolutionized outcomes in several different tumor types but is relatively unsuccessful in treating ER+ breast cancer. Our project aims to dissect the immune microenvironment through multipronged approaches for assessing the type and function of infiltrating immune cell subsets in treatment-naive tumor tissue and peripheral blood from patients with ER+ invasive ductal carcinoma (IDC) and invasive lobular carcinoma (ILC), the two major histological subtypes of ER+ breast cancer. Through comprehensive global analysis of immune cells and their potential interactions, we identified macrophages and CD8 T cells as important subsets that can be potentially targeted in ER+ breast cancer to improve patient outcomes.

Objective:
- The ongoing COVID-19 pandemic, caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), led to more than 126,265,724 million infected individuals and over 2,769,853 global deaths by the end of March 2021. The novel coronavirus SARS-CoV-2 is an enveloped, positive-sense, single-stranded RNA betacoronavirus of the family Coronaviridae. The main target of these virus is the lungs, but new reports about COVID-19 indicate its effects on other organs, including kidney, brain and testis. SARS-CoV-2 uses the angiotensin-converting enzyme 2 (ACE2) receptor and the transmembrane protease serine 2 (TMPRSS2) to invade human cells. Based on ACE2 and TMPRSS2 expression, the testis has been classified as being at high risk of infection by SARS-CoV-2. Indeed, patients with COVID-19 suffer from testicular discomfort, which can be associated with viral orchitis. Similarly, some studies have reported reduced testosterone levels in the serum from COVID-19 patients. These data suggest that COVID-19 could affect male reproductive function, in fact the presence of SARS-CoV-2 has been detected in the testis of some deceased patients of COVID-19 as well as in some semen samples of patients in the acute and recovery phase of COVID-19. Our preliminary analysis of the testis of 5 patients deceased due to COVID-19 and their respective controls showed increased germ cell apoptosis, micro thrombosis, and infiltration of lymphocyte CD3+. Strikingly, we observed that almost all the apoptotic cells were spermatogonial stem cells (SSCs). Non-apoptotic SSCs also displayed signs of DNA damage and oxidative stress. Using single nuclei RNA-seq, RNAscope, and immunohistochemistry, we will interrogate the effects of COVID-19 on human testis from the macroscopic presentation to the single cell, providing an important basis to understand the SSC alterations that may hamper fertility in COVID-19 survivors.

The Cytometry Facility is proud to announce the Cytoflex LX is on the way! For our last virtual seminar, we will be re-introducing the technology. In brief, the CytoFLEX LX Flow Cytometer is an expansion of the CytoFLEX Platform. This flagship model expands research possibilities with six lasers and 21 color parameters. Six spatially separated lasers allow panels to be spread across the spectrum reducing cross talk and spectral overlap. The fully activated instrument includes three fluorescent channels from the 355 nm (UV) laser, five from the 405 nm (Violet) laser, three from the 488 nm (Blue) laser, five from the 561 nm (Yellow Green) laser, three from the 638 nm (Red) laser, and two from the 808 nm (Infrared) laser. The available 96-well plate sample loader allows for high-throughput applications and fits inside the instrument preserving the compact footprint. This event will start at 9:00 a.m. with numerous talks from trainees and conclude with a keynote lecture at 12:30 p.m.
Our Keynote speaker is Dr. Graham Walker, American Cancer Society Professor and HHMI Investigator at MIT, and a member of the National Academy of Sciences. He will deliver a talk from 12:30 to 1:30 p.m. entitled “Unravelling Mutagenic Translesion Synthesis: A Journey from the Ames Test to Cancer Chemotherapy,” and will meet with trainees afterward. Dr. Walker holds an R35 NIEHS merit award entitled, “Mechanisms of Eukaryotic Environmental Mutagenesis” and is researching therapeutics that inhibit mutagenic translesion DNA synthesis to sensitize cancer cells to chemotherapy.
